Fibroblasts Drive Metabolic Reprogramming in Pacemaker Cardiomyocytes
The sinoatrial node (SAN) initiates all heartbeats within the heart. The diseases of SAN result in catastrophic consequences from bradycardia to cardiac arrest. The sinoatrial node (SAN) is characterized by the microenvironment of pacemaker cardiomyocytes (PCs) encased with fibroblasts. A unique self-truncation of bacterial GH5 endoglucanases leads to enhanced activity and thermostability
GsCelA is a 368-amino acid endoglucanase secreted by the thermophilic bacterium Geobacillus sp. 70PC53 isolated from a rice straw compost in southern Taiwan. This enzyme belongs to the glycoside hydrolase family 5 (GH5) EGs with a TIM-barrel structure that is common among this type of enzymes. The purified recombinant GsCelA displays unique self-truncation process, resulting in the removal of 53 amino-acid peptide from the C-terminus, leading to higher specific activity and thermostability than the original full-length enzyme. A similar self-truncation/activation process has been observed in at least two other GH5 EGs produced by Gram-positive bacteria. Unveiling the secret of coral associated microbial aggregates
Microbial aggregates can be found in many different animals and plants. These aggregates play important roles in their host with various ecological functions, like symbiosis or parasitism. Although several reports have claimed the finding of microbial aggregates reside in corals; however, the knowledge of their physiology, biochemistry, morphology, genetics, ecological functions… is barely studied.
A hybrid mRNA vaccine induced broadly neutralizing antibodies against Omicron and other SARS-CoV-2 variants
Wuhan wild-type strain-based vaccines became almost ineffective against new SARS-CoV-2 variants. Academia Sinica mRNA Alpha team constructed a hybrid mRNA vaccine containing mutations of Omicron and Delta variants. When used as a booster in mice previously immunized with wild-type vaccine, the hybrid vaccine induced high titers of broadly neutralizing antibodies against all tested variants, as well as T cell responses against conserved T cell epitope. This hybrid vaccine also includes important mutation sites of Omicron BA.4/5, its effectiveness against the new dominant strains is being explored......
