Activation of GABA transmission by clonazepam reverses the autistic-like phenotypes of the Cav3.2 knockout mice

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by social and communication deficits, accompanied by restricted and repetitive behaviors. Our findings suggest that loss-of-function in the Cav3.2 channel contributes to ASD-like phenotypes through disrupted GABAergic signaling and that pharmacological enhancement of GABAergic signaling may offer a potential therapeutic approach for individuals with ASD carrying the CACNA1H mutations.

Equilibrative nucleoside transporter 2 modulates inosine catabolism to influence astrocyte metabolism and reactivity

Our findings demonstrate that inosine is a vital fuel for astrocytes under inflammatory stress and that Ent2 is the key transporter controlling astrocytes to utilize this fuel.

Dendritic cell-targeted liposomes for cancer immunotherapy via inhibition of aryl hydrocarbon receptor

Ligands for the aryl hydrocarbon receptor (AhR), such as kynurenine derived from the tumor microenvironment, are well-known immunosuppressants.Through specific interaction between SP65 and CD11c, CH223191 is specifically transported to DCs. This inhibits AhR function, promoting IL-12 and IFN-γ production in DCs and NK cells, respectively.

Population-specific polygenic risk scores for people of Han Chinese ancestry

The Taiwan Precision Medicine Initiative recruited more than half a million Taiwanese residents, providing a large dataset of genetic profiles and electronic medical record data for people with Han Chinese ancestry. Using extensive phenotypic data, we conducted comprehensive genomic analyses across the medical phenome with individuals genetically similar to Han Chinese reference populations.

The Taiwan Precision Medicine Initiative provides a cohort for large-scale studies

Han Chinese comprise nearly 20% of the global population yet remain underrepresented in genetic studies 1,2, creating an urgent need for large-scale cohorts to advance precision medicine.

Dimer dissociation and aggregation hot-spot exposure synergistically accelerate light chain variable domain aggregation associated with AL amyloidosis

Light chain (AL) amyloidosis is a life-threatening systemic disorder caused by the aggregation and deposition of antibody light chain (LC) fragments in multiple organs, including the heart and kidneys.

Structural basis of enhanced stalling efficiency of an engineered ribosome arrest peptide

In this study, we investigated the molecular mechanisms of ribosome stalling induced by an engineered ribosomal arrest peptide (eRAP).

Structural basis of K11/K48-branched ubiquitin chain recognition by the human 26S proteasome

We report cryo-EM structures of human 26S proteasome in a complex with a K11/K48-branched Ub chain. The structures revealed a multivalent substrate recognition mechanism involving a hitherto unknown K11-linked Ub binding site at the groove formed by RPN2 and RPN10 in addition to the canonical K48-linkage binding site formed by RPN10 and RPT4/5 coiled-coil.

The Drosophila ZER1 homolog interacts with ref(2)P to regulate autophagy and Keap1-cnc/NFE2L2/Nrf2-mediated oxidative stress

The ubiquitin-proteasome system (UPS) and macroautophagy/autophagy are two major pathways for maintaining cellular protein homeostasis. Increasing evidence has highlighted the complex interactions and crosstalk between these pathways; however, the specific molecules and mechanisms mediating the interplay between the UPS and autophagy are still not fully elucidated.

Environment-dependent mutualism–parasitism transitions in the incipient symbiosis between Tetrahymena utriculariae and Micractinium tetrahymenae

Our results demonstrate that incipient endosymbioses employ mitochondrial remodeling and endosymbiont metabolic reprogramming to actively regulate transitions between mutualistic and parasitic states, revealing how symbiotic partnerships navigate environmental pressures during their incipient stage of evolutionary establishment.

Fibrillarin Methylates a Movement Protein to Help Satellite RNA Trafficking in Plants

Here, we show that the arginine-rich motif (ARM) of P20 directs its nucleolar localization, enabling its interaction with FIB in the nucleolus and Cajal bodies, where FIB methylates the ARM.

In a new study, an international team of researchers has discovered a remarkable strategy in which stinkbugs form a defensive alliance with fungi, using them to physically protect their eggs from wasp parasitism.

Hepatitis B surface antigen level identifies patients with inactive chronic hepatitis B from Asia with HCC risk below surveillance threshold

Chronic hepatitis B (CHB) is a major global health concern primarily due to hepatocellular carcinoma (HCC) development.Serum HBsAg levels <100 IU/mL effectively identify patients with inactive CHB with negligible HCC risk and limited viral activity, which is important in optimising surveillance strategies and defining partial HBV cure.
